Droplet-based single-cell RNA-sequencing struggles with the samples that matter most in translational research: the rare, precious, and low-input populations of a few hundred cells. PB10X, a plate-based method built on Smart-seq3xpress and made fully compatible with 10X Genomics 5' chemistry, resolves these samples one individually verified cell at a time.

This application note describes how the PB10X workflow can run on the F.SIGHT for image-indexed, monoclonality-documented single-cell isolation and the I.DOT for non-contact nanoliter reagent dispensing.
